Adaptation of in vitro regeneration protocol for Brazilian wheat genotypes Ciência Rural

ABSTRACT: The availability of an efficient protocol for in vitro regeneration is imperative for genetic transformation. Using genotypes adapted to the target region as a transgenic platform accelerates the development of cultivars. Therefore, this study aimed to adapt an in vitro regeneration protocol for Brazilian wheat genotypes. For this purpose, the in vitro regeneration capacity of immature embryos from six Brazilian wheat genotypes using two protocols of regeneration of somatic embryos was analysed. Furthermore, combinations of 2,4-D and picloram in the callus induction medium were tested in order to improve regeneration efficiency. Androgenic response of wheat genotypes resistant to fusariosis PAB

Abstract: The objective of this work was to assess the androgenic response, via microspore culture, of wheat genotypes with different levels of resistance to Gibberella zeae. The number of androgenic embryos per spike, and of green and albino plants was counted for the BRS 179 (moderately resistant), Frontana and Sumai 3 (resistant), and BRS 194, Embrapa 27, and Fielder (susceptible) genotypes. The degree of interference by the Fielder, Pavon 76, and Sumai 3 ovary-donor genotypes, used for co-culture with the microspore cells, was also assessed regarding androgenic response. O morangueiro é uma espécie multiplicada vegetativamente, o que facilita a progressão de viroses a partir de mudas infectadas. A utilização de matrizes provenientes do processo de cultivo in vitro tem sido uma prática na obtenção de mudas isentas de patógenos. Diversos protocolos de micropropagação recomendam a utilização de quatro subcultivos, para preservar a identidade genética dos clones e manter alta taxa de multiplicação. ABSTRACT: Gene stacking refers to the introduction of two or more transgenes of agronomic interest in the same plant. The main methods for genetically engineering plants with gene stacking involve (i) the simultaneous introduction, by the co-transformation process, and (ii) the sequential introduction of genes using the re-transformation processes or the sexual crossing between separate transgenic events. In general, the choice of the best method varies according to the species of interest and the availability of genetic constructions and preexisting transgenic events. Impact of high-molecular-weight glutenin alleles on wheat technological quality PAB

Abstract: The objective of this work was to determine high-molecular-weight glutenin subunits (HMW-GS) and their relationship with technological quality parameters in a collection of wheat (Triticum aestivum) genotypes. Two hundred and seventy-two accessions were evaluated on SDS-Page, and molecular markers were used to identify the 7oe allele and 1BL.1RS translocation. For 219 accessions with a homogenous glutenin profile, 53 profiles and 21 alleles were identified. The most frequent combination was 2*/7+9/5+10 (11.9%). The mean value of genetic diversity for the three assessed Glu-1 loci was 0.67. Indução de embriogênese somática em diferentes explantes de aveia (Avena sativa L.) Ciência Rural

Três experimentos foram conduzidos com o objetivo de avaliar o potencial de formação de calos embriogênicos de três tipos de explantes de aveia branca (Avena sativa L.) (embrião imaturo, embrião maturo e segmentos de coleóptilo). Cada experimento foi conduzido com um dos explantes, cinco cultivares ("UPF15", "UPF16", "UPF18", "UPFA20" e "OR3") e dois meios de indução de calos (M1 = MS + 2mg L-1 de 2,4-D e M2 = MS + 2mg L-1 2,4-D + 1mg L-1 BAP), sendo avaliadas as freqüências de calos embriogênicos aos 60 dias de cultivo, massa fresca (mg) aos 90 e 120 dias e taxa de crescimento dos mesmos durante um período de 30 dias. Phenotypic variability in a population of globe artichoke Ciência Rural

The existence of variability is a prerequisite for genetic improvement in plants. Globe artichoke is a high nutritious vegetable with medical value, representing a profitable alternative for rural producers. This research was conducted with the aim of evaluating the phenotypic variability in a commercial cultivar of artichoke (Cynara cardunculus var. scolymus L) established from seeds. Field plants were assessed when primary head reached commercial stage. An amount of 21 quantitative and 5 multicategoric characters were assessed. This article discusses, from the standpoint of cellular biology, the deterministic and indeterministic androgenesis theories. The role of the vacuole and of various types of stresses on deviation of the microspore from normal development and the point where androgenetic competence is acquired are examined. Based on extensive literature review and data on wheat studies from our laboratory, a model for androgenetic capacity of pollen grain is proposed.
